1520 Tissue mechanics driven symmetry breaking and cellular reprogramming during regenerative wound healing

Symmetry breaking initiates tissue regeneration after injury. Both chemical and mechanical factors contribute to constitutes the optimal microenvironment, known as morphogenetic field, for cellular reprogramming occur allow regenerative wound healing. Here, using single-cell sequencing, CellChat epigenetic sequencing technologies, we demonstrated that an optimized environment not only enhances wounding-induced hair neogenesis (WIHN) but also facilitated chromatin remodeling, endogenous reprogramming, dermal-epidermal signaling dermal condensation during WIHN. Specifically, softening bed via Blebbistatin treatment decreased collagen/integrin expression, increased Wnt signaling, expanded area of Axin/Wnt/β-catenin expression number condensations in bed, implying expansion competent zone regeneration. Based on this, propose a model with 4-element loop, which force, cell behavior, fate, microenvironment co-regulate each other. Optimized mechanics favors by decreasing cell-matrix binding increasing cell-cell adhesion force. Driven motility (force/energy), new primordia form. Thus, have further delineated chemo-mechanical, symmetry events WIHN identified evo-devo advantages fostering stiffness observed African spiny mice.
